G'day Gazz, so many ways to skin this cat, opinions aplenty, truth is there is no real right and wrong as it's what's right for you that matters with one qualifier, the parts have to work together to get good results.
Based on what you have listed and not trying to change your mind on the cam this is what I think.
The S&S barrels and pistons are good stuff, other option is to have your cylinders bored to match a set of good pistons.
TB, the S&S is good, so is the SE and HPI better again, price point might influence you there.
    S&S lifters are really good.
Pushrods, pick one, I use the SE tapereds, many years, zero issues.
585 is a good mid range cam, set at 10.5 or thereabouts static with a good pipe it goes hard.
Heads is where I would say send yours out to a good headporter and get them matched to you cam and TB, set the combustion chamber CC's to match your pistons for the compression you are after, get comp releases while you are at it.
That's a basic outline anyway, you can go the full blueprint route, it will give a bit more, for a street riden Harley only you can decide if the cost is worth the gain.
I'm sure you will get a bit of interest here, lots of fella's with heaps of experience pop in from time to time.
You said you had a builder in mind, have you confered with him on your plan?

Your builder is the one to talk to mate, have trust in him, tell him what your end expectations are and be guided by his decision and hope you can
    afford it.
